Why the loading dose of Aspirin is 325 mg in ACS?

Experimentally, a single oral 100-mg dose of aspirin is sufficient to completely block the synthesis of thromboxane A2, the predominant pathway by which aspirin inhibits platelet aggregation. Studies comparing the dose effect of aspirin noted increased bleeding complications with higher doses while observing no differences in efficacy.

Newer studies suggest that 162 mg may be as effective as and safer than 325 mg for the acute treatment of STEMI.
